The goal of my research is to create simulated particle holograms for use in the analysis of Holographic Microscopy data. Holographic Microscopy is a tool that is used to analyze objects on the micrometer scale, especially in drug development. I based my holograms on the Rayleigh Scattering equations, a set of equations used to estimate the path of scattered light.
Holographic microscopy is a particle imaging technique with many scientific applications. Like all imaging techniques, it is susceptible to aberration – the distortion of a beam of light caused when it passes through a lens. The Grier lab at NYU uses Lorenz-Mie scattering equations to analyze data to correct distortions produced by the lenses in holographic microscopes. This paper outlines the process of creating a simulation and visualization of Rayleigh Scattering – a simplified alternative to Mie scattering in Python to achieve a better understanding of the phenomenon. Physics texts and code modules are used to research the concepts and math behind the theory. The simulation is coded in Python, using Matplotlib for the visualization aspect.
