Protocol

1.  Standard SHIELD protocol.

2.  Passive delipidation with Clear+ Delipidation Buffer.

3.  Passive labeling using 2 μL DCS2 in 5 mL of PBST for 3 days at 37°C.

4.  EasyIndex index matching and mounting. 

5. Imaged on SmartSPIM with 3.6X objective.
