Measure ~0.0500 g of turmeric.
Place turmeric and 5.00 mL of ethanol into a test tube and mix throughly.
Let sample extract for 5.00 min. Mix occasionally.
Filter the extract to remove the leftover turmeric powder.
To create a filter paper cone, fold it in half twice (in fourths overall), and open one of the folds (if you open directly in the middle, there will be a gap in your filter).
Place the filter paper cone in the funnel.
Pour the extract mixture into the filter.
Compare your extract with the calibration solutions. Dilute if necessary.
Blank (0 μM), curcumin calibration solutions (5, 10, 15, 20, and 40 μM), and the undiluted extract (Ex)
Note that the extract is much more intense in color than the calibration solutions. The solution is too concentrated and will block much of the available light from reaching the detector in the spectrometer.
Dilute the extract with ethanol until the color is less intense in comparison with the standard sample.
Start with a 1:10 dilution.
undiluted extract in test tube
draw up 1.00 mL of extract with
1-mL volumetric pipet
transfer 1.00 mL of extract to 10-mL graduated cylinder
dilute with ethanol to 10-mL mark; mix thoroughly
Compare the 1:10 diluted extract to the calibration solutions.
curcumin calibration solutions and undiluted extract
1:10 dilution
The 1:10 dilution of the extract is still too concentrated.
Dilute again.
1:10 dilution in graduated cylinder (center)
draw up 1.00 mL of the 1:10 dilution with 1-mL volumetric pipet
transfer to 10-mL graduated cylinder (right)
dilute with ethanol to 10-mL mark (right); mix thoroughly
Compare the 1:100 diluted extract to the calibration solutions.
curcumin calibration solutions and undiluted extract
1:100 dilution
The 1:100 diluted extract is dilute enough to fit with the calibration solutions.
Obtain the spectrum of the 1:100 dilution.
