Inoculating From a Slant
To inoculate from a slant, do the following:
Follow steps 1-13 on the Aseptic Transfers page.
Insert your inoculating tool into the tube and quench.
When quenching an inoculating tool in a slant, place the tool where the agar and tube meet.
3. Touch your tool gently to the organismal growth. You do not need a large amount of inoculum unless specifically instructed.
4. Remove your tool from the culture tube.
5. Flame the lip of the tube and replace the cap.

To inoculate a slant, do the following:
Remove the cap of your tube and flame the lip.
Insert your inoculating tool into the tube. Trace a “fishtail” pattern on the surface of the slant. [NOTE: You will inoculate some slants, such as TSI, in a different manner.]
Remove your tool from the tube. Flame the lip and replace the cap.
Flame your tool.
