To inoculate a deep, do the following:
Flame your needle.
Remove the cap of your tube and flame the lip.
Insert your needle straight down into the deep and remove it by pulling it straight up. How deep you insert the needle will depend on the test. It is difficult to make a perfect stab, but it is possible to read all the tests we do with an imperfect stab.
The deep on the left illustrates the needle stabbing directly down into the agar. The deep on the right shows a stab that sliced through the agar. You are, however, able to read the results using either tube
4. Flame the lip of the tube and replace the cap.
5. Flame your needle.
