Forebrain
Forebrain organoids are generated from iPS cells. Is a serum-free 4 stages protocol, including embryoid bodies formation, neuroepithelial expansion, differentiation and maturation up to 8-9 months. During the organoid development you can face the expression of the main developmental brain markers like PAX6, SOX2, p-Vimentin, as well as the markers for the intermediate zone (TBR2+), and cortical plate (CTIP2, MAP2, TBR1). Midbrain
We generate human midbrain organoids from neural precursor cells activating the Wnt and Shh pathway by small molecules. The protocol is a 5 stages protocol, beginning with embryoid body formation step followed by expansion of neuroepithelia, prepatterning, patterning and maturation stages. Midbrain organoids asume their maturation starting from 40 up to 120 days in culture including dopaminergic neurons. [Price inquire]
Neuro-immune Assembloids
We generate neuro-immune assembloids combining functional iPSC-derived microglia in brain organoids. NeuroI-Assembloids can be generated combining organoids and microglia from the same iPSC lilne or from two different lines (e.g. pathological vs healthy). Blood Brain Barrier (BBB)
We propose a BBB model on chip, using the exclusive system from Netri. The Duplex system is especially designed for co-culture of 2 different cell types by membrane. Each chip is compartmentalized by membrane technology. This enables co-culture of the brain organoids with a combination of endothelial cells, astrocytes and pericytes. Up to 16 data points, Duplex is composed of 2 compartments (1 channel (IR and OR) and 1 open well) for independent culture per chips and used the membrane technology to compartmentalize culture and connect channel to open well. Duplex is available in NeoBento™ a standard ANSI format (96-well plate), pump-free & expensive equipment-free and compatible with standard equipment (liquid handling & imaging) and readouts (IF, ELISA, Lysis cells analysis, Liquid chromatography…).
