Cerebral Organoid differentiation of ES or iPS cells
Cerebral organoids are generated by using Self organizing, single-rosette cortical organoids (SOSR-COs) protocol (PMC10724074). The protocol begins with dual-SMAD inhibition on monolayer of pluripotent stem cells. On day 4, the monolayer cut into small square of similar size and replate on 100% geltrex layer to form 3D structures. On day 10, these SOSRs individually remove from the geltrex by using Stripper micropipetter and plate in ultra low attachment U-bottom 96 well plate. These organoids will be culture in 96 well plate till day 30. Organoids are transfer to ultra low attachment 24 well plate to deliver to the investigator on day 30.
