PEI

DAY BEFORE

- plate Hela cells in complete medium (without antibiotics)

10cm-dish: 2x10⁶ cells

1x6well: 3x10⁵ cells

DAY OF

- Prepare fresh 1 mg/mL PEI solution in sterile H2O (NB: Always use fresh PEI !)

- Prepare transfection mix:

recipe for 1 mL (1x6well = 1 mL; 10cm dish = 6 mL)

2 µg of DNA

20 µL of PEI solution (must be 10 fold the DNA)

4 µL of PBS 10x

Add water to 40 µL total

- Incubate 20 min RT

- Add 1 ml of complete medium and add to cells (without washing)

- Change medium after 5-6 h 37°C.
