Several pharmaceutical solvent systems commonly employed by the pharmacist during the extemporaneous dispensing of minoxidil topical solution using Loniten tablets were evaluated. These included ethanol, propylene glycol, water and a commercially available liquid, Vehicle/N. A total of twenty-five (10 mg) Loniten tablets were pulverized in a wedgewood mortar for five minutes. The tablet powder was then dissolved in the experimental solvent system and stirred thoroughly for ten minutes. The liquid dispersion was filtered using the vacuum filter method and the powder residue was then washed twice to obtain a fixed volume of the filtrate. The concentration of minoxidil in each solvent system was determined spectrophotometrically at the wavelength range of 280 to 282 nm depending upon the solvent. The amount of minoxidil extracted in each sample was calculated using the previously constructed standard curve in the respective solvent system. From the data obtained, it was concluded that Vehicle/N is the optimum solvent for such use, and the drug recovery was about 99%. Whereas, the mixture of methanol/propylene glycol/water at a volume ratio of 6:2:2 was observed to be the second best with a drug recovery of 97%. The addition of sodium lauryl sulfate had no effect in enhancing the solubility of drug when used in a solvent with poor recovery profile of drug. Also, the presence of excipients of the tablet dosage form did not interfere with the extraction process of minoxidil.

The aim of this work is to apply the melt crystallization technology to manufacture ibuprofen tablets coated with polyethylene glycol in a single step. This technology, based on a pastillation process, allows in situ separation between two components (active ingredient and coating material). The design and application of this technique depend on the thermo-physical properties of the substances used, as well as on the existence of a eutectic point in the phase diagram. To evaluate the prerequisite conditions, first, DSC curves, allowing the construction of the phase diagram of the binary system, were investigated and the eutectic point was determined (30 mass% ibuprofen, 52 C). Then, the stability of the selected mixture (10:90 mass% of ibuprofen, PEG6000) was studied by thermogravimetric analysis. Finally, the coating quality was investigated under different operating conditions including viscosity, cooling plate temperature, the power of ultrasound and seeding. This parametric study showed that seeding with PEG6000 is necessary to obtain a hemispherical pastille shape, a suitable separation and a pure and thick coating layer. In addition to the optimization of operating conditions of the in situ coating process, it was possible to determine the optimum viscosity and the cooling plate temperature (271.77 m Pa s, 25 C) to obtain a uniform and crystalline coating. During the deposition of molten drops on the cooled surface, the progression of crystal growth was monitored online by optical microscopy. According to the good separation achieved and to the purity and thickness of the microscopic cross-sectional material, the in situ coating process is conceivable for the production of PEG6000-coated ibuprofen tablets.

Glibenclamide is a second generation sulfonyl ureas drug that are widely used in diabetes type 2 therapy. Glibenclamide in floating tablet formulations is used to improve the bioavailability. The aim of this study was to determine the optimum composition of floating tablets of glibenclamide by simplex lattice design. The optimum formula was determined based on numeric method with physical properties, floating characters, and dissolution profile of tablets. the results of the study show that glibenclamide floating tablets have good physicochemical properties. The combination of HPMC K100LV and Carbopol 940 decreases floating lag time and dissolution rate of floating glibenclamide tablets. The optimum composition of glibenclamide floating tablet was HPMC K100LV 44.29 mg and Carbopol 940 15.71 mg with desirability value 0.989.
