We study protein homeostasis system in bacteria.
Proteins are vital catalysts for all living organisms. The maintenance of protein quality control in bacteria is directly associated with their survival in various natural environments and host niches. Therefore, diverse host immune system and man-made antimicrobial substances target bacterial protein homeostasis. However, the protein quality control system has been poorly understood in the pathogens and commensal strains, since the study has been mainly limited to laboratory Escherichia coli strains.
The Microbial Genetics Lab investigates bacterial protein quality control systems using diverse bacterial models, including the major opportunistic human pathogen Pseudomonas aeruginosa, Pseudomonas putida, which is widely used in bioremediation and biotechnology, the cold-adapted bacterium Pseudomonas antarctica, and commensal Escherichia coli strains. We employ a multidisciplinary approach integrating microbial genetics, genomics, proteomics, and protein biochemistry to elucidate the molecular mechanisms underlying bacterial proteostasis, environmental adaptation, and survival.
Our research will provide a concrete foundation to understand how bacteria have evolved to survive various exogenous and endogenous proteotoxic stresses. The results of this work will be useful in the development of anti-microbial/anti-virulence agents that target bacterial protein homeostasis as well as the promotion of beneficial gut commensal bacteria.

What organisms do we use?
We use Pseudomonas aeruginosa as a model for bacterial pathogenesis and Escherichia coli as a model for studying the gut microbiome. We also investigate other Pseudomonas species, including Pseudomonas putida, which is widely used in biotechnology, and Pseudomonas antarctica, a cold-adapted bacterium.
We also use the eukaryotic model organism Saccharomyces cerevisiae (budding yeast), particularly as a humanized yeast model to investigate molecular mechanisms associated with human diseases.
We use Caenorhabditis elegans (a nematode) and human cell lines as host models to investigate host–microbe interactions.

What is the role of chaperones in protein homeostasis?
Molecular chaperones play a central role in maintaining cellular protein homeostasis (proteostasis) by assisting proper protein folding, preventing stress-induced protein aggregation, and facilitating the recovery and recycling of misfolded or aggregated proteins. Our laboratory investigates the functions and molecular mechanisms of chaperone systems using an integrated approach combining genetics, physiology, biochemistry, and omics-based analyses.
