We integrate molecular optogenetics, mRNA optogenetics, and RNA biology to build a frontier‑level technology portfolio. Our goal is to translate these platforms into RNA editor–based and optogenetic therapeutics through strategies that are precise, reversible, and programmable.

● Molecular Optogenetics: Light‑precision control of molecules inside living cells and brains
We pioneered molecular optogenetics, moving beyond channelrhodopsin‑based neuronal depolarization to directly switch DNA, mRNA, and protein functions on and off with light. With LARIAT (Nature Methods, 2014), we introduced the first method to reversibly trap target proteins in light‑induced clusters, enabling ultrafast, high‑resolution control of cell signaling. We then showed with OptoSTIM1 (Nature Biotechnology, 2015) that endogenous calcium channels can be controlled in vivo to enhance memory in mice. Most recently, SynapShot (Nature Methods, 2024) allowed real‑time, 4D tracking of synapse formation and elimination in the live mouse brain, visualizing the molecular remodeling of synapses during learning and movement. These advances extend the concept of “optogenetic medicine” to disease models—including Parkinson’s disease, depression, and dementia—and are underpinned by 30+ representative publications, patents, and technology transfers.

● RNA Control & Editing: DNA‑sparing next‑generation therapeutics
We advance CRISPR‑Cas13 platforms that precisely cleave, modify, and regulate RNA—overcoming limitations of DNA editing. Photo‑activatable Cas13 (Nature Communications, 2024) links Cas13 to a light‑responsive module so base editing occurs only upon illumination, achieving spatiotemporally selective RNA editing with minimized off‑target effects in cells and animals. In parallel, programmable RNA acetylation (Nature Chemical Biology, 2024) equips Cas13 with catalytic enzymes to chemically modulate RNA function, establishing a first‑in‑class platform for functional RNA remodeling. With mRNA‑LARIAT (Nature Cell Biology, 2020), we optically controlled the localization and translation of endogenous β‑actin mRNA, defining a causal link between local mRNA translation, cell morphology, and plasticity. Together, these tools enable precision control, reduced side effects, and patient‑specific applications for RNA medicines.

● RNA Optogenetics (New RNA Biology): A fused paradigm for light‑switchable RNA
By combining the spatiotemporal precision of optogenetics with the target specificity of RNA editors, we are establishing “RNA optogenetics”—a new paradigm in which RNA behavior is turned on and off by light, and further tuned by chemical signals. This fusion yields reversible, cell‑type– and time‑specific RNA interventions that naturally extend into high‑precision RNA therapeutics. Beyond basic discovery, this strategy positions our work to drive Optogenetic Medicine 2.0 and RNA Editing as two pillars of next‑generation biotechnology, with strong potential to shape the future bio‑therapeutics market.
