  • Post-doctoral Fellow (m/f) in crystallography at SNBL’s BM01

Context & Job description The Swiss Norwegian Beam Lines at the European Synchrotron Radiation facility (SNBL at ESRF) provide scientists from both Norway and Switzerland with access to synchrotron radiation on two beamlines BM01 and BM31. SNBL operates around the clock state-of-the-art, custom-designed instrumentation for x-ray scattering and absorption experiments. The successful candidate will complement a group of scientists operating the beam lines. Within this team, the candidate will participate in the operation of SNBL’s crystallography and material science diffraction station BM01. The beamline performs a variety of diffraction experiments with single crystals, powders, and thin films, as a function of temperature, pressure, external fields, including in-situ and operando conditions. Your mission includes: Supporting visiting user teams as a local contact Develop his/her own research program Contribute to the continuous technical upgrades and development of the beamline in collaboration with other staff members Expected profile: PhD in chemistry, or physics, or materials science Experience in crystallography, powder diffraction.   • To start on February 1st 2021 as a permanent assignment, we are looking for an experienced small molecule crystallographer, with knowledge of X-ray devices, synchrotron facilities and with a passion for promoting the electron diffractometer in the fields of science and industry. Last year, Dr. Gustavo Santiso-Quinones, Dr. Gunther Steinfeld, Dr. Eric Hovestreydt and Nils Gebhardt founded the start-up company ELDICO Scientific. ELDICO stands for the Electron Diffraction Company.It emerged from a ground-breaking scientific publication on electron diffraction, which was nominated as Breakthrough of the Year by SCIENCE magazine in 2018. Electron diffraction and the electron diffractometer constitute a disruptive technology that will fundamentally change the way research is conducted at the nano-crystalline level.   • The European Synchrotron ESRF in Grenoble, France, will resume its activities this year after a complete machine upgrade, which will bring exciting new possibilities of experiments, in particular in time-resolved serial synchrotron crystallography (SSX). The Structural Biology Group is currently building a beamline dedicated to SSX planned to perform time-resolved experiments with a microsecond time resolution. In order to prepare these experiments, we are developing an offline setup to perform microsecond time-resolved UV-vis absorption spectroscopy experiments on protein crystals. This time-resolved setup is part of the in crystallo optical spectroscopy (icOS) Lab, (formerly known as the Cryobench , http://www.esrf.eu/home/UsersAndScience/Experiments/MX/icOS.html ), which provides various optical spectroscopic techniques (UV-vis absorption, fluorescence, Raman) that can be applied to proteins in the crystalline or solution state, before, during, or after an X-Ray diffraction experiment. We are looking for a postdoc interested in contributing to the development of the TR-icOS setup and to associated scientific projects coupling time-resolved optical spectroscopy and X-ray crystallography. The successful candidate should have strong expertise in at least one of the following topics: optical spectroscopy, instrumentation, X-ray crystallography. Integrative structural biology and biophysics to analyze and design

RNA interactions of RNA‐Recognition Motif (RRM) proteins

ESR5 project: RRM structure and design for synthetic biology and bio‐analytics applications

This project will design RRM proteins with novel RNA binding properties and develop small molecules to

modulate RNA binding activity. The design will be confirmed using experimental structural biology. The

modulation of RNA binding by small molecules will be experimentally assessed by combining state‐of‐the‐

art solution NMR and biophysical techniques (ITC, FP, SPR). The project is in collaboration with

computational groups of the ITN for the design of RRM variants and for experimental validation in cellular

assays.

ESR6 project: Conformational dynamics and RNA binding specificity of RRMs using integrative structural

biology and biophysics

This project will characterize the dynamics and RNA recognition of RRMs using a combination of advanced

solution NMR (relaxation measurements to characterize internal dynamics from nanosecond to

millisecond timescales) and biophysical techniques (ITC, FP, SPR). These experiments will identify the role

of specific residues and dynamic features in RRM proteins for RNA binding specificity and identify key

residues and molecular features involved. In close collaboration with computational groups of the ITN,

RRM variants that modulate RNA recognition will be designed and experimentally verified.

ΠΕΡΙΓΡΑΦΗ ΘΕΣΗΣ Η εξοικονόμηση ενέργειας και βελτίωση της ενεργειακής αποδοτικότητας, όπως για παράδειγμα στα δίκτυα μεταφοράς ρεύματος, βασίζεται στην ανάπτυξη νέων τεχνολογιών (π.χ. ηλιακές κυψελίδες, υπεραγώγιμα καλώδια, κινητήρες υψηλής απόδοσης κ.α.). Αυτές απαιτούν την κατανόηση εξωτικών καταστάσεων της ύλης, στις οποίες αναδύονται και γίνονται εμφανείς οι κβαντικές ιδιότητες των ατόμων. Τέτοια ασυνήθιστη συμπεριφορά εκδηλώνεται στους υπεραγωγούς, υλικά τα οποία αφήνουν το ηλεκτρικό ρεύμα να περνά με πρακτικά μηδενική αντίσταση. Το ερευνητικό πεδίο της διδακτορικής διατριβής αφορά στην κατανόηση της φυσικής νέων δισδιάστατων κρυσταλλικών ενώσεων (π.χ. αλογονίδια ή χαλκογονίδια) μεταβατικών μετάλλων για εφαρμογές χαμηλής ή μηδενικής κατανάλωσης ενέργειας. Για το σκοπό αυτό ο υποψήφιος διδάκτωρ θα εστιάσει στη δημιουργία καινοτόμων φυσικών υβριδικών υλικών που εκμεταλλεύονται τις «διεπαφές» μεταξύ μαγνητικών/ μεταλλικών / μοριακών νανομετρικών στρωμάτων. Θα εκπαιδευτείτε σε καινοτόμες πειραματικές μεθόδους (π.χ. σκέδαση ακτίνων-Χ και νετρονίων, μαγνητομετρία SQUID κλπ) ώστε να διευρύνετε την επιστημονική σας γνώση μέσω της μελέτης της δομής και δυναμικής των υλικών αυτών. Development of ultrastable fluorescent proteins for quantitative super resolution microscopy at cryogenic temperature.

He/she will investigate the structural photophysics of fluorescent proteins used as markers in super-resolution microscopy in conditions of cryogenic temperature.

The ultimate goal will be to engineer a variant that efficiently photoactivates at such low temperature.

Candidates should have a strong background in biophysics and/or biochemistry. Preliminary experience in protein crystallography, molecular biology and/or advanced optical microscopy will be key advantages.

  • Postdoc: Layered Metal Chalcogenides (magnets and superconductors)

The Institute of Electronic Structure and Laser (IESL) of the Foundation for Research and Technology - Hellas (FORTH), in the frame of the project "Deciphering the workings of molecule intercalated iron chalcogenides" (DOMINION), funded under the Office of Naval Research (ONR) Global (ONR Announcement #N00014-17-S-B001), is seeking to recruit one (1) post-doctoral researcher.

Job Description

DOMINION provides an intimate engagement of syntheses and characterisation for a wide range of layered iron-based chalcogenide compounds. The successful post-doctoral candidate will focus on the fundamental physics and chemistry of inter-atomic interactions of solid-state iron-chalcogenide magnets and superconductors, made up of functional molecular components with nanoscale dimensions. For this, user-facility photon and neutron science US DOE labs, with their advanced analytical instrumentation, optimized for high -flux/ -resolution/ -data throughput, unavailable anywhere else in the world, will be utilized to generate key insights on how (structural and electronic / magnetic) correlations at the atomic level of matter are built and influence the superconducting electrons to pair-up. Knowledge of physical crystallography, including diffraction experiments and data analysis strategies, will be highly valuable for the proposed research endeavors.

  • We have an opening for a highly motivated scientist in the Synchrotron Crystallography team at the EMBL Grenoble Outstation.

This team works in close collaboration with the ESRF Structural Biology Group in the operation and development of high performance structural biology beamlines with the aim of impacting on challenging structural biological problems. Towards this goal, as well as providing world leading experimental facilities, the EMBL-ESRF Joint Structural Biology Group also develops advanced instrumentation and data collection methods.

The successful candidate will contribute to the operation and development of methods for biological micro-crystallography on the ESRF beamlines ID23-2 and/or ID29. The recent upgrade of ID23-2 to a high intensity microfocus beam (~2 um in diameter), and installation of a high performance MD3-Up diffracometer represents an excellent opportunity to further develop techniques in biological micro-

crystallography. This position also allows the opportunity to contribute in the complete upgrade of ID29 to provide an extremely high intensity, tunable, and microfocus beamline for serial crystallography data collection. He/She is expected to actively participate in local and international initiatives aimed at the further development of synchrotron serial and micro-crystallography techniques required to maximise the scientific potential of ID23-2 and ID29 post ESRF Extremely Brilliant Source upgrade.

The successful candidate should have a PhD degree in Structural Biology or a related discipline and a minimum three years of post-doctoral experience in MX data collection and analyses. A practical knowledge, or an interest, in synchrotron beamline instrumentation and developing new methods for micro-crystallography at advanced light sources is essential. The candidate should be familiar with the latest MX software developments. Experience in computer programming would be advantageous.

  • The Müller group at EMBL Heidelberg in collaboration with GlaxoSmithKline (GSK) seeks to recruit an outstanding postdoctoral fellow to explore the use of cryo-EM to study small-molecule ligand-bound structures of chromatin complexes. The successful candidate will apply recent cryo-EM methodological developments to study epigenetic targets; develop protocols that increase throughput and support lead optimization in drug discovery projects; acquire high resolution data sets of ligand-bound chromatin complexes and other epigenetic targets.

The successful candidate holds a Ph.D. in structural biology or biochemistry. The project is particularly suitable for a postdoctoral fellow with solid previous experience in cryo-EM who is interested in utilizing and further developing cryo-EM methodology for studying epigenetic targets and visualizing small molecule ligands in the drug discovery process. The position requires exceptional motivation and commitment, the ability to work independently as well as in a team and excellent technical, organizational and management skills. Συνδυάζοντας δομικά συστατικά με αντικαρκινική δράση, με άλλα που αυξάνουν την διάρκεια κυκλοφορίας νανοσωματιδών στο αίμα, ΘΑ ΑΝΑΠΤΥΧΤΟΎΝ ΠΡΟΗΓΜΈΝΕΣ (ΝΑΝΟ)ΜΟΡΦΈΣ ΧΟΡΉΓΗΣΗΣ χημειοθεραπευτικών φαρμάκων. A 2-year Post-Doctoral position is available immediately in the Laboratory of Molecular Biology and Genomics in the Department of Biochemistry and Biotechnology of the University of Thessaly, located in the city of Larissa. The selected candidate will work in the framework of a national Research Infrastructure program on Synthetic Biology.

Research in the Laboratory of Molecular Biology and Genomics focuses on various aspects of insect olfaction, reproduction and evolution, aiming at the development of new approaches for control of insects of agricultural or medical importance. We are seeking a highly motivated and creative scientist with a track record of successful research. A good bioinformatics background is strongly desired. The ideal candidate should be able to analyze raw sequencing data, perform genome assembly and annotation and have a good understanding of comparative and functional genomics. At the same time, he/she should be able to design and perform experiments in support of the bioinformatics analyses.

to support a structure-based drug discovery project on various GTPase families (headed by Dr. Oliver Daumke). The successful candidate will also be involved in the operation of the BESSY II bio-macromolecular crystallography beamlines (headed by Dr. Manfred S. Weiss) and will take part in graduate student education.

Initially, a three-year contract will be offered to the successful candidate with the possibility for an extension. The salary will be based on German Federal TVöD. The position is available from March, 2018.

The appointed Group Leader will be an ambitious structural or structural-cell biologist with an original research programme on structure-function relationships of macromolecular complexes in eukaryotic systems. He/she will benefit from the world-class environment offered by the inter-institutional Partnership for Structural Biology (EMBL, ESRF, ILL, IBS, http://www.psb-grenoble.fr) which provides access to state-of-the-art instrumentation including the recently upgraded ESRF synchrotron X-ray beamlines for MX and SAXS, cryo-EM (Polara/K2, Titan Krios/K2), high-field NMR (600-950 MHz spectrometers), the ILL neutron scattering beamlines and a biophysics platform. He/she will also have direct access to in-house bacterial, insect and mammalian cell expression facilities, confocal microscopy and high-throughput crystallization platforms for soluble and membrane proteins. We are particularly interested in candidates who will make use of local opportunities in advanced X-ray crystallography and cryo-EM and/or are interested in structural-cell biology e.g. cryo-electron tomography, super-resolution microscopy and correlative light and electron microscopy. Existing groups at EMBL Grenoble focus their research on protein-nucleic acid complexes involved in transcription, epigenetics, RNA metabolism, RNA virus replication and host-pathogen interactions.


Applicants should have a PhD, at least 3 years post-doctoral experience and a strong record of achievement in structural, molecular or cell biology. An initial contract of 5 years will be offered to the successful candidate. This is

Postdoctoral fellow

The positions are available starting January 2018. We seek highly motivated individuals with an excellent track record and experience in protein purification and biophysical and biochemical characterization of proteins and protein complexes, preferably including membrane proteins.   • Position: Postdoctoral Fellow in Integrated Structural Biology in the Viral Infection and Cancer (VIC) Group at the Institut de Biologie Structurale (IBS) under the supervision of Dr. Joanna Timmins

Title: Study of the hNTH1/YB1 Complex: a new anti-cancer drug target

Project summary: The most prevalent non-surgical cancer treatments function by generating DNA damage. Such therapies generate dose-limiting toxicities in normal tissues, but are often efficacious since cancer cells often display impaired DNA repair systems and proliferate more rapidly than most normal cells. Nonetheless, in such cells, functional DNA repair pathways provide a common mechanism for cancer-therapy resistance. The development of resistance to cytotoxic therapy that was initially effective is extremely common and is a major clinical issue. Drugs targeting repair pathways thus exhibit a promising potential as new anti-cancer agents. This project aims at characterising a complex formed between a human DNA repair enzyme (the DNA glycosylase hNTH1) and a transcription factor (Y-box binding protein, YB1) that is implicated in resistance of tumour cells to the chemotherapeutic drug, cisplatin. More specifically, our objectives are to: (i) determine the structures of hNTH1 alone and in complex with either lesion-containing DNA or YB1, (ii) characterise the N-terminal extension of hNTH1 and its role in regulating DNA repair activity and interaction with YB1 and (iii) gain further structural information regarding YB1. Structural information will be obtained using a combination of X-ray crystallography, small-angle X-ray scattering and NMR.

The project is funded by the French Labex GRAL and involves the collaboration between three partners: the teams of J. Timmins and M. Blackledge at IBS and the CMBA screening platform at CEA/BIG Grenoble.

Laboratory: The IBS is located on the European Photon Campus (alongside EMBL, ILL and ESRF) in Grenoble, France. The IBS performs interdisciplinary research at the interface of biology, physics and chemistry and benefits of a privileged access to the EMBL High Throughput Crystallization (HTX) platform to explore crystallization conditions and to the Synchrotron Radiation source, ESRF, to collect diffraction data.

Qualification requirements: The applicant should hold a PhD degree in Structural Biology or in Biophysics obtained in 2015, 2016 or 2017. Prior experience with X-ray crystallography and/or NMR would be a strong asset.

Terms of employment: 18 months contract with the Université Grenoble Alpes. Gross salary ~2500€/month.

  • Research engineer permanent position in Macromolecular Crystallography Application deadline: June 30th, 2017 The Institut Pasteur in Paris is one of the leading institutes worldwide in fundamental biology and biomedical research. It is home to more than 130 research groups with scientists from over 70 countries. The recently created Center for Innovation and Technological Research (Citech) includes all Core Facilities of the Institut Pasteur, including the X-Ray Crystallography Platform (PFX). The PFX has two major missions: (1) to provide research teams working in the field of macromolecular crystallography with the expertise and technology for high-throughput crystallization, X-ray diffraction measurements and crystallographic computing, and (2) to offer expertise in bio-crystallography and structural analysis to research groups in biology at Institut Pasteur and outside organisations. The PFX is looking for a research engineer in Macromolecular Crystallography, who should fulfill the missions and tasks described thereafter under the supervision of the head of the facility.   • PhD (CIFRE) position – SANOFI & ESRF on cryoEM methods applied to antibodies

PhD thesis title: High resolution Cryo- Electron Microscopy of Antibody / Antigen complexes

More and more research focus is being put on biologics and their development as drugs. These molecules are usually large and flexible and they can interact with various targets located on the surface of cells; Bi-specific antibodies are an example that can potentially bind to two different antigens and which have gained attention due to their potential application in cancer immunotherapy and drug delivery. To fully understand their mechanism, structural knowledge at an atomic level is needed.

While low resolution studies can be obtained using traditional microscopy, high resolution studies of these interactions are usually performed by protein crystallography (or by Hydrogen Deuterium Exchange studies) using fragments of both antibody and antigen. The ESRF is in progress of installing a Krios microscope which, coupled with a direct electron detector and a Volta phase plate will enable us to determine the full structure of the complexes.

The successful candidate will characterise Ag/Biologics complexes from Sanofi, using state-of-the-art microscopes at the cryo-EM platform of the Partnership for Structural Biology (PSB) and will determine the high-resolution structure of one or more of these complexes.

The PhD thesis will last 3 years starting in late autumn 2017, in a collaboration between Sanofi (Vitry sur Seine, France) and the ESRF (Grenoble, France) under the co-supervision of Dr. Magali Mathieu (Sanofi) and Dr. Gordon Leonard (ESRF). This thesis will be CIFRE (Conventions Industrielles de Formation par la REcherche) and the student will be a Sanofi employee (CDD) for the whole duration of the thesis. Most of his/her work will be performed at the ESRF.

The applicant must have a Master Degree (Master2 or MSc) in integrated structural biology or biophysics, some knowledge in cryo-electron microscopy and in molecular and cell biology. He/she should be a team player and have good communication skills.

  • A position is available for a Professor of Structural Biology at the University of St Andrews.

Structural biology in St Andrews is based in a large shared purpose-built laboratory, capable of hosting ~40 researchers. This space is currently shared with the Gloster, Schwarz-Linek, Taylor and Pliotas groups. The laboratory has great facilities for protein biochemistry (incubators, floor centrifuges), crystallisation (Gryphon crystallisation robot for dispensing nano-drops and Scorpion robot for mixing crystallisation screens), crystal visualisation (two crystallisation incubators and Minstrel robot for automated crystal imaging), and in-house crystal testing (Rigaku generator with two CCD detectors; one robotically controlled).

Crystallographers have regular access to BAG synchrotron time, both at Diamond and ESRF, which is shared with users in Dundee. St Andrews provides an excellent mass spectrometry/proteomics facility for protein identification and more complex analyses. Solution state biomolecular NMR is supported by a dedicated 700 MHz instrument (Bruker Ascend, Prodigy TCI cryoprobe). Structural biology is also supported by a uniquely powerful EPR setup (X, Q, W band); St Andrews hosts the two most sensitive pulse EPR spectrometers in the UK. A new biophysics suite provides ITC (Microcal PEAQ and VP), SPR (Biacore T200), CD, and stopped-flow machines.

There are also dedicated rooms for insect and mammalian cell culture, with hoods and large incubators containing shaking platforms for protein expression.
