From its establishment, the parish was under the administration of diocesan priests from the Archdiocese of Manila. It was then transferred to the Diocese of Lipa after it became a suffragan of Manila in 1920. It remained a part of the Diocese of Lipa until November 28, 1966 when the Holy Father Pope Paul VI created the Diocese of San Pablo through the apostolic letter Eccleasiarum per ampla. From the canonical erection of the diocese in April 18, 1967 until now, the parish is under the Diocese of San Pablo.
The Most Rev. Pedro N. Bantigue, D.D. served as the first bishop of the diocese from 1967-1995. He was followed by the following:
Bishop Francisco C. San Diego. D.D. from 1995 until he was transferred to the new Diocese of Pasig in 2003.
Bishop Leo M. Drona, S.D.B., D.D. who served from 2004 until his retirement in 2013.
Bishop Buenaventura M. Famadico, D.D. who served from 2013 until his retirement in 2023. Msgr. Mylo Hubert C. Vergara, D.D., Bishop of Pasig, became the Apostolic Administrator until November 2024.
Bishop Marcelino Antonio M. Maralit, D.D. who took possession of the Cathedral on November 21, 2024.
There are no remaining records of the first parish priests from 1815 until 1895 for the first two Books of Baptism no longer exists.
