blaIMP-1によってコードされる伝達性カルバペネム耐性遺伝子は、1988年に富山県の病院から分離された緑膿菌GN17203ではじめて報告され、その後、blaIMP-1はエンテロバクター属を含む他の細菌でも検出されるようになりました。現在、多くのblaIMPのバリアントが存在しており、また、blaIMPプロモーターの点変異がプロモーター強度を変化させることが報告されております。緑膿菌GN17203で最初に報告されたblaIMP-1のプロモーター(Pc)は、発現強度が低いプロモーター(PcW)でした。本研究ではプロモーター領域の点変異は、抗菌薬選択圧のもとで強力なプロモーターの方向へとシフトしていくのかを調べました。バイオインフォマティクスの手法を用いてPseudomonadota門の14,529のゲノムデータから115のblaIMPを抽出しマルチプルアラインメント解析を行いました。抽出した115個のblaIMPのプロモーター解析の結果、そのほとんどがクラス1インテグロンに位置するPcを利用していました(n = 112, 97.4%)。年代別ごとにプロモーターを解析した結果、強力なプロモーターであるPcSを持つblaIMP集団の出現は一過性であることが明らかになりました。一方、PcWにTGn拡張型の-10モチーフを獲得した中程度のプロモーター強度を持つPcW-TG集団は、徐々に世界中に広がっていることが明らかになりました。Pcプロモーター強度とIntl1インテグラーゼの切断効率のあいだには逆相関があることが報告されております(切断効率が落ちるとblaIMPの伝播効率が低下します)。このようなトレードオフの関係から強力なプロモーターを持つblaIMPが急速に拡散する可能性は低いということが示唆されました。その一方で、他のインテグロンを利用することでプロモーター強度が増加する可能性は否定できません。プロモーター解析を含むblaIMP遺伝子のモニタリングは今後も必要であると考えております。
