As cell culture efforts get more streamlined and produce more protein, there becomes a need for ways to purify cell culture material into therapeutic antibodies. The final design developed consists of two protocols that can be used to eke out more protein binding in the Protein A Affinity chromatography step of antibody purification. The first protocol optimizes protein binding by running the mobile phase at lower and lower flow rates as more and more protein binds to the column. The second protocol optimizes protein elution by running an elution buffer only up until the point where all protein on the column elutes off. Future testing on chromatography optimization could look at other forms of chromatography such as ion exchange and hydrophobic interaction columns.
