COLONY PCR (S. cerevisiae)

• Use pipette tips instead of toothpicks (picks fewer cells), to place yeast in 24 μl of a 3 mg/ml zymolyase solution, in a PCR tube.

• Incubate at 37 C for 30 min.

• Add 26 μl ddH2O.

• Follow the colony PCR protocol for E. coli (i.e., boil for 10 min and then PCR, as described for the E. coli colony PCR here).
