Course summary
Day 1 and 2 or 3.
Basics on protein structure. Protein purification and chromatographic resins and strategies (NTA, Blue Sepharose, hydroxyapatite, ionic anionic exchangers, hydrophobic binding, size exclusion). Selection of proteins to be purified in the course and team development. SDS Page and protein quantification. Protein expression using vectors.
Day 3.
In silico analysis of protein physical-chemical properties of targeted proteins, selection of a purification strategy. Preparation of chromatographic material
Day 4-7.
Protein puriļ¬cation and characterization experiment timeline (4 days). The students should be able to purify the selected overexpressed protein in the following 4 days.
Day 8.
Spectroscopical analysis of purified proteins (Absorbance and fluorescence)
Day 9.
Feed-back and roll up.
