Method to sterilize equipment and media
Media recipes
These are the most basic media recipes. There are an infinite number of media recipes.
For all recipes:
Sterilize at 121°C for at least 15 minutes. Allow to cool a bit before pouring media.
Recipes are calculated for 1 L of media.
The % agar can be changed. All of the recipes below are at 2%.
Similarly, you can play with the concentration of sugars either increasing or decreasing the carbon source.
Distilled water can be used in place of deionized water.
For sporulation of Fungi
- Water agar
20 g agar
1 liter deionized water
- Oatmeal media
25 g oatmeal
20 g agar
Cook oatmeal in 500 mL of DI water at 50-60°C for 30 to 45 minutes, stirring every few minutes. Strain through a fine sieve or cheese cloth. Bring up the sieved liquid up to 1 L with deionized water. Add agar.
For growth of Fungi
- 2% malt extract agar (MEA)
20 g malt extract
20 g agar
1 L deionized water
- Potato dextrose broth agar
Method 1
25 g potato dextrose broth
20 g agar
1 L DI water
Method 2
200 g of potatoes; peeled, washed, and diced
1 L of water
20 g agar
Add the pieces to 1L of deionized and boul for 20 to 25 minutes. Remove the potato pieces with a fine sieve or cheesecloth. Add the agar to the sieved liquid and bring up to 1 L.
