All of known life uses the process of transcription to produce messenger RNA from a DNA template. A grand challenge is to understand how transcriptional output depends on regulatory DNA sequence and cellular context. I will introduce the field of transcriptional regulation, describe progress on this grand challenge and discuss the role(s) that mathematics will need to play. I will include a combination of concrete results and conceptual musings.
