Project Title:

Optimization of Agrobacterium mediated transformation protocol for local elite cultivars of Foxtail millet (Setaria italica).

Objectives:

1. To develop tissue culture regeneration protocol for high frequency shoot regeneration through somatic embryogenesis.
2. To develop a stable gene transfer protocol through Agrobacterium mediated transformation
3. Molecular analysis of transgenic Setaria italica lines

Progress made:

• The calli induced from 2,4 D (2.0 mg/L) regenerated with BAP (1.5 mg/L) showed differentiation frequency (82%).
• The regeneration frequency is more in MS media containing BAP (1.5 mg/L).
• Rooted plants will be grown to maturity in the greenhouse and allowed to set seeds and the work is in progress
