Identified the approximate shape of the outer seal. It can be approximately the diameter of a champagne bottle cork. It was also suggested to buy a butt-plug because it does essentially everything we need minus the access hole for the colonoscope.

There's an inflatable enema device which can be used to model a balloon design to inflate and seal/stay inside a person's sphincter.

There's a material called Ecoflex that is soft and stretchy and could be used to make a balloon.
We can use PDMS for the seal.
No two batches of PDMS will come out the same because each batch needs to crosslink and depending on the ratio, temperature, rpessure, environment the PDMS is casted in, all the properties is subject to change. So the online properties of PDMS won't be applicable for every batch of PDMS. It's similar to polyester resin where the cure time and structural integrity depends on all of these factors.

Apparently, the device doesn't have to be made out of FDA approved material for human testing as long as we do the NSR right, or the form Dr. Kalmaz gets signed.
