In collaboration with Doug Young, also in Chemistry at W&M, we recently published a study of simple diynes and their effects on bacterial cell cultures. We synthesize primary and secondary polyynes in my lab on larger scale (ca. 0.1 to 1.0 grams) and the biological testing is done in Prof. Young's lab.
An important note is that some of these polyynes exhibit selective action against pre-formed biofilms. We continue to make new compounds and evaluate their effects on bacterial cultures.
Bacterial viability assay for the propargylic diyne compound array. Compounds screened for 10 hours at a 5 mg/mL concentration in E. coli. Ampicillin, a previously demonstrated antibiotic was used as a viability control, and DMSO was employed to demonstrate the small percentage of DMSO utilized to prepare stocks was not affecting bacterial growth.
