Skills and Techniques
Microbiological techniques: Generation of knockouts by homologous recombination in Candida, Knockout screening in Candida, Gene deletion strategy, Bacterial and yeast transformation.
Bacterial cell culture: E. coli- (DH-5α and Rosetta), Making competent and ultra-competent E. coli cells, Blue-white screening, Spotting assay, Replica plating.
Molecular techniques: Molecular Cloning [Restriction-free (RF) cloning, Site-Directed Mutagenesis (SDM)], SDS-PAGE, Western blotting, DNA extraction and purification, Agarose gel electrophoresis, PCR, Colony PCR, Plasmid PCR, Fusion PCR, Overlap extension PCR.
Protein purification: Ni-NTA chromatography, Gel filtration chromatography/Size exclusion chromatography, Pull-down (Immunoprecipitation) using lipid-coated agarose beads, Protein preparation using TCA method.
Mammalian Cell Culture: Maintaining cell lines, freezing, storage, transfection, producing pseudoviruses (HIV core pseudoviruses), harvesting pseudoviruses, expressing non-classical amino acid in pseudoviruses using orthogonal translation system.
Bioinformatics and statistical analysis: Sequence alignment (NCBI-BLAST), Primer design, ExPASy's ProtParam tool, T-COFFEE, MATLAB, PyMol, ChimeraX, SnapGene.
