Open valve to CO2 tank and cool stage until completely frozen.
Cut straight across the cerebellum so brain can stay level and upright.
Apply 1% ethanol to frozen stage as the base.
When EtOH is halfway frozen, add a layer of 30% sucrose to the base.
When sucrose is halfway frozen, place brain so that the dorsal side will face towards the blade.
Add more sucrose around the tissue and allow to freeze.
Manually lower or raise the stage so that the tissue is below where the blade will run.
Place blade in its designated space and tighten screws to hold the blade in place.
Obtain a brush to handle the tissue and sectioned dish full of distilled H2O to place tissue.
Begin cutting tissue into sections of the desired thickness (50 microns) and fully extend cutting arm for automatic adjustments.
When done cutting, remove blade wash with soap and water, then dry. Coat blade in oil and store until next use.
For mounting, place one section in dish of water, float to the top and place slide under tissue in water.
Using a brush, guide the tissue to its spot on the slide.
When done mounting, allow your tissue to dry before staining.
Start out with six containers. For the first, add distilled water. For the second, add 70% alcohol. For the third, add absolute alcohol. For the fourth, add clear safe. For the fifth, add cresyl violet stain. The sixth is left empty behind the others.
Take slides, make sure back half of glass is facing out, and put in container with distilled water. Carefully place in slots and leave for 2 minutes.
Take slides out of distilled water and put in 70% alcohol. Leave in for 2-4 minutes.
Take slides from 70% alcohol and place in absolute alcohol. Leave in for 5 minutes.
Move slides from absolute alcohol and place in clear safe. If a smoke-like appearance occurs in liquid, the sections haven't been properly dehydrated and needs to be put back in the alcohol. Leave in for 5 minutes or until tissue is fully cleared.
The tissue, which is slightly more transparent, is taken out of the clear safe and placed in the absolute alcohol. Leave in for 4 minutes.
Take slides out of absolute alcohol and place in 70% alcohol. Leave in for 4 minutes.
Takes slides out of 70% alcohol and place in distilled water. Leave in for 2 minutes.
Take slides out of distilled water and place in cresyl violet stain. Leave in for 10 minutes, but person from video suggests 20 minutes.
Add acid alcohol to sixth container and put container between the distilled water and 70% alcohol containers.
After 20 minutes, move the slides out of the cresyl violet stain and place in the distilled water. Water starts to change color, dump it out in waste container and refill. Leave it in a little longer.
Take slides out of distilled water and place in acid alcohol. The acid alcohol will then change color, dump it out and refill. Do this quickly so not to dehydrate tissue.
Take slides out of acid alcohol and place in 70% alcohol. Leave in for as long as you see necessary (eyeball it to see how much tissue is still stained).
Take slides out of 70% alcohol and place into absolute alcohol. Let it sit for a little bit longer.
Take slides out of absolute alcohol and place in clear safe. Leave these here for a while, at which point it should be safe and won't be losing stain.
Prepare cover slips by taking off gloves. Carefully grip edges to separate the two slips. A rainbow sheen means more than one is there, wiggle a bit to make sure there is only one. Three slides mean three cover slips, the cover slips should be laid sticking out, so they are easily grabbed.
Open paramount and use one-sided Q-Tip to get it by placing inside the paramount like a stick.
Carefully remove slide from the clear safe and make sure the side with the brains are facing upwards. Hold it at angle facing away from you and take Q-Tip and apply paramount to bottom edge of slide facing away from you.
Take cover slip and place perpendicular to the slide about half-way down the cover slip and moving it up at a decreasing angle. Continue moving until it's flat with the slide. Tap on each slide to make sure it's even with both edges.
Do this with the remaining slides and leave out on a flat surface to dry.
