Liquid biopsy profiling of PARPi-resistance breast cancer patients indicates that many patients develop multiple reversion mutations, and that alterations in the 53BP1-Shieldin pathway are present but less frequent. The co-occurrence of 53BP1-Shieldin pathway mutations and reversion mutations suggests parallel mechanisms of resistance can operate in the same individual with metastatic disease.

Our study dissects the interplay between telomerase and ALT pathways of telomere maintenence in cancer cells. We provide evidence of the incompatibility between telomerase RNA subunit - hTR and the recombination‐mediated ALT mechanism of telomere elongation, through its non‐canonical functions.

We identified ATRX/DAXX histone chaperone complex as a novel physical and functional interactor of the central FA pathway member, FANCD2.ATRX/DAXX complex cooperates with FANCD2 and the MRN exonuclease at sites of stalled replication forks to support the deposition of histone variants H3.1 and H3.3, the recruitment of the key HR factor CtIP, and ultimately the HR-mediated restart of the stalled replication forks.

We identified distinct models of FA pathway function during DNA ICL repair vs. the restart of an APH-stalled replication fork. Our study reveals a clear separation of function within the 3-tiered FA pathway and crucial functions of FANCD2 at stalled replication forks that is completely independent of the upstream FA core complex.
