If a Knight is dead, but the King/Knight win condition is met, does the dead Knight win too?

Yes.

If the Assassins are all dead, but the King dies without another win condition being met, do the Assassins still win?

Yes.

If I play a card that says someone "loses the game", how does that work?

When a player loses the game by any means, if it would fulfill a win condition, the game would end and the winners would be declared based the rules for winning the game. Otherwise that player would be killed and the game would continue on.
