Abstract: Cancers progress often involves the reactivation of embryonic programs through expression of transcription and epigenetic factors. In aggressive hepatocellular carcinoma (HCC), embryonic factors, such as Alpha-fetoprotein (AFP), are used diagnostically to monitor tumour burden. However, whether this represents an entire embryonic program and how it is activated is unclear. We have examined how transcriptional and epigenetic networks are regulated in hepatocyte differentiation using human pluripotent stem cells. Recently, we used single cell Multiomics to compare normal liver and matched adjacent non-tumour and HCC samples. We compared networks involved in hepatic differentiation with changes associated with HCC to determine the similarities and differences between cancer and embryonic development.
