To test our hypothesis, we will first use CRISPR/dCas9 to knockdown expression of PA14_54520 in the PA14 strain. We will then test whether a reduction in expression of PA14_54520 confers any growth defect when the bacteria is grown in media that contains the tricarboxylate cis-aconitate or in artificial urine media.
