With the stroke it makes the lines of your design thicker without having to draw multiple lines to make it thicker. The fill helps with filling in a spot quickly, and not having to drawing for like 30 minutes 

Some other things that are important is naming your layers, because it helps with organizing your stuff. Naming your layers helps with placing stuff together, like all the left eye go together, and everything for the right eye goes in the same layer. See how laying things together can help? But you can also not laying things together if you want that specific piece to do something different from everything else.
