Library Preparation

When requesting a run on the Illumina high-throughput sequencer, you agree to submit samples that meet the following requirements:

  • Genomic DNA prep: DNA should be intact and high quality, gel photo is required. A minimum of 5 ug with a concentration of 200-500 ng/ul in TE. (If you used a nanodrop to check the concentration please provide a concentration in the range of 400-1000 ng/ul, because the nanodrop does not measure genomic DNA accurately.)
  • ChIP-seq prep: 10 ng of ChIP enriched DNA in 30 ul of water (in a low-retention tube)
  • Pre-made Libraries: A minimum of 20 ul of 10 nM pre-made library in EB (Tris-Cl 10mM, pH 8.5) must be provided in a low retention tube. A gel photo is required indicating size of library.
  • Custom Primer: Custom sequencing primer concentration should be at 100 uM, with a minimum volume of 20 ul.
