Please discuss your requirements with the manager prior to ordering conjugations.
What do I need to provide?
Please provide us with 100ug of purified antibody (IgG from any species) in PBS with no carrier protein (eg BSA, low % glycerol is acceptable but it can disrupt the post conjugation quantification). This is the minimum we recommend to conjugate but we can conjugate as little as 50ug.
We suggest the antibodies are validated for flow cytometry or at least IHC prior to conjugation. A protocol for validating antibodies will be posted on this web page in the near future.
Purified antibodies can be obtained from Biolegend, R&D Systems, ThermoFisher and BD Biosciences. However other vendors will provide them. They must be carrier free preparations.
How much antibody can be recovered?
Typically we would expect to recover 60-70% of the labeled antibody following the conjugation reaction. This will be provided in ~200uL of stabilising buffer.
What information regarding the reaction do I get?
We will provide you with the antibody concentration post conjugation and also a mass spect trace of the antibodies to show conjugation was successful.
How many tests will in-house conjugations provide?
This is hard to assess. We recommend you perform a titration on an appropriate sample but as a guide we suggest using 2uL of labelled antibody in a 100uL reaction volume. A protocol for titrating antibodies will be posted on this web page in the near future.
How much does it cost?
Conjugations are charged at £120/antibody to University of Manchester researchers. External researchers should contact the facility for a quote.
