Can CD4⁺ cytotoxic T cells kill as effectively as CD8⁺ cytotoxic T cells?
Our study provides clear experimental evidence that they can.
Published in @ScienceAdvances, the study shows in humans that CD4-CTLs are as potent killers as CD8-CTLs. Using an integrated multi-omics approach, the team identifies a rare stem cell memory CD4-CTL subset (TSCM-CTLs)—pre-committed to the cytotoxic lineage and potentially crucial for resolving viral infections.
Importantly, the researchers also establish an in vitro differentiation model to generate durable, long-lived, functional CD4-CTLs, opening exciting avenues for immunotherapies, cell-based therapies, vaccine development, and precision medicine.
This work strongly advocates exploring both CD4-CTLs and CD8-CTLs in next-generation vaccines and immunotherapies.
