Since the discovery of tyrosine phosphorylation being a critical modulator of cancer signaling, proteins regulating phosphotyrosine levels in cells have fast become targets of therapeutic intervention. The nonreceptor protein tyrosine phosphatase (PTP) coded by the PTPN11 gene "SHP2" integrates phosphotyrosine signaling from growth factor receptors into the RAS/RAF/ERK pathway and is centrally positioned in processes regulating cell development and oncogenic transformation. Dysregulation of SHP2 expression or activity is linked to tumorigenesis and developmental defects. Even as a compelling anti-cancer target, SHP2 was considered "undruggable" for a long time owing to its conserved catalytic PTP domain that evaded drug development. Recently, SHP2 has risen from the "undruggable curse" with the discovery of small molecules that manipulate its intrinsic allostery for effective inhibition. SHP2's unique domain arrangement and conformation(s) allow for a truly novel paradigm of inhibitor development relying on skillful targeting of noncatalytic sites on proteins. In this review we summarize the biological functions, signaling properties, structural attributes, allostery and inhibitors of SHP2.

It's time to set sail! Even if you live nowhere near a lake or ocean, you will get to do some sailing in this science activity as you build your own toy sailboat. But first, you have to make sure your boat doesn't capsize! Are you up for the challenge?

But boats don't just need to float - they also need to stay upright and avoid capsizing (flipping over). To do this, they need a low center of mass, meaning their weight is concentrated towards the bottom of the boat, not the top. That might seem like it's a problem for sailboats, boats with very tall sails that stick way up into the air. How do they stay balanced with so much mass concentrated way up high? They do so with another part called the keel, which is on the bottom of the boat (so if you've only seen a sailboat from above the water, you might not even know the keel exists!). The keel is a big part under the boat, shaped like a fin, that serves two purposes. It holds the ballast, or heavy weight, that helps lower the boat's center of mass. It also helps prevent the boat from being blown sideways by the wind. In this project you made a keel from nails and aluminum foil, which helped prevent your sailboat from flipping over and helped it go straight.

US Sailing Certified Level 1 Small Boat Instructors guide campers during week-long Sail Camps in June and July using Optimist Prams and Sunfish sailboats. Our Sail Camps are intended to introduce newer sailors to the sport or help older campers brush up on the skills they learned last summer in a safe and fun environment. Water safety and good sportsmanship are essential in all of our Sail Camp activities! All campers will need to be able to swim without fear of the water. We will play sailing games, learn knots, and practice the basics of sailing (capsize recovery, points of sail, rigging, how to notice weather and wind direction, sailing terminology, sail trim, and how to skipper and crew a dinghy). Sailing: We teach beginner and intermediate sailing skills in our Sunfish fleet using the US Sailing curriculum. All participants will be required to learn capsize recovery and demonstrate the skill during their first lesson. A sailing workshop series taught by women, for women. Come learn sailing basics without intimidation! Ladies 18 years and older will sail in our Sunfish fleet. This series will consist of 10.5 hours of instruction led over the course of three days (Fri 5PM - 730PM, Sat 8AM - 12PM, Sun 1PM - 5PM).
