Many cellular processes in cancer are attributed to kinase signaling networks. V-akt murine thymoma viral oncogene homolog (AKT) plays a major role in the PI3K/AKT signaling pathways. AKT is activated by PI3K or phosphoinositide-dependent kinases (PDK) as well as growth factors, inflammation, and DNA damage. Signal transduction occurs through downstream effectors such as mTOR, glycogen synthase kinase 3 beta (GSK3), or forkhead box protein O1 (FOXO1). The abnormal overexpression or activation of AKT has been observed in many cancers, including ovarian, lung, and pancreatic cancers, and is associated with increased cancer cell proliferation and survival. Therefore, targeting AKT could provide an important approach for cancer prevention and therapy. In this review, we discuss the rationale for targeting AKT and also provide details regarding synthetic and natural AKT-targeting compounds and their associated studies.

Since debut, CLASS:y have set their sights on both Korea and Japan, signed under Universal Music for the latter. They remain on the periphery of both markets, though their music is slowly improving. Dave Grohl wrote and recorded the song in its original form in 1991. Grohl played the demo of the song for Nirvana band-mate Kurt Cobain in a break between touring for the album Nevermind. According to Grohl, "I'd told him I was recording and he said, 'Oh, I wanna hear it, bring it by.' He was sitting in the bath-tub with a walkman on, listening to the song, and when the tape ended, he took the headphones off and kissed me and said, 'Oh, finally, now I don't have to be the only songwriter in the band!' I said, 'No, no, no, I think we're doing just fine with your songs.'"[4] Nirvana would jam the song on soundchecks during the Europe 1991 tour.

The song was a setlist staple from 1995 to 2000. It disappeared afterward and wasn't played again until the Echoes, Silence, Patience & Grace tour in 2007, where it was played twice. The song would not be played again until 2012 on the Wasting Light tour, where it was again played twice. The song was also played twice on the Sonic Highways tour in 2015. It was most recently played at a secret show in 2018.[5]

With the exception of the also brilliant C&C Thang, this song feels the most out of place in the entire game. Yet Target (Mechanical Man) is one of the first of many songs by Klepacki that ultimately directed the eclectic nature of the C&C franchise, and heavily influenced what was to come.

Many artists tell me that their song is intended for their own recording project, but they would be thrilled if other artists recorded it. There are indeed instances where a song that is well-suited for its writer can work well for additional artists. The focus of the song is rightly on the targeting of kids. This will resonate will most parents, whatever their racial and social background. I assume gender woo is deep within US schools. It is good to see a high profile and catchy pushback. There is about to be a generation of detransitioners. The debate needs to happen, both about the transing of children and the support this receives from the corporate world. 


You are right about Chinese factories, but it is whataboutery in that transing of children is happening now in the US including in poorer communities, and also needs to be addressed.

They're also rapping about God taking revenge and in the video they show only rainbow-coloured products, not the tucking and binding garments. I'm pretty sure the creation of this song was driven by the homophobia that's so common amongst the Americochristian right. Sorry but I don't see these rapture rapsters as allies.

Mutations in gene regulatory elements have been associated with a wide range of complex neuropsychiatric disorders. However, due to their cell-type specificity and difficulties in characterizing their regulatory targets, the ability to identify causal genetic variants has remained limited. To address these constraints, we perform an integrative analysis of chromatin interactions, open chromatin regions and transcriptomes using promoter capture Hi-C, assay for transposase-accessible chromatin with high-throughput sequencing (ATAC-seq) and RNA sequencing, respectively, in four functionally distinct neural cell types: induced pluripotent stem cell (iPSC)-induced excitatory neurons and lower motor neurons, iPSC-derived hippocampal dentate gyrus-like neurons and primary astrocytes. We identify hundreds of thousands of long-range cis-interactions between promoters and distal promoter-interacting regions, enabling us to link regulatory elements to their target genes and reveal putative processes that are dysregulated in disease. Finally, we validate several promoter-interacting regions by using clustered regularly interspaced short palindromic repeats (CRISPR) techniques in human excitatory neurons, demonstrating that CDK5RAP3, STRAP and DRD2 are transcriptionally regulated by physically linked enhancers.
