Structural biology of proteins associated with Fatty acid biosynthesis of Mycobacterium tuberculosis by using NMR, X-Ray crystallography, Small Angle X-Ray Scattering (SAXS) and Single-particle Cryo-electron microscopy. Studying the interaction between AcpM and dehydratase genes along with studying the effect of thiourea compounds against dehydratase genes of MTB FAS pathway. Studying the mechanism of substrate selectivity of monooxygenase genes of MTB FAS pathway. The answer to these questions will help in designing anti-TB drugs which target these steps of MTB FAS pathway and will help in the treatment of TB, which is a global health concern due to the emergence of antibiotic- resistant strains.
Techniques to be used:
Cloning, Expression and Purification of proteins
Crystallization, X-ray Crystallography, Nuclear Magnetic Resonance, Small Angle X-ray Scattering
Drug inactivation assay, Cell cytotoxicity assay
Single molecule Cryo-electron microscopy
