The use of microscopes is a critical skill in A-level Practical. You are required to know how to:
prepare the specimen,
mount it on a slide,
stain [sometimes],
observe and
draw the visible biological structures.
Below, we shall learn skills 1 - 4. Please read the following and practice often.
1. Preparing the specimen
(a) Cutting specimen (such as onion, potato)
(b) Obtaining inner epidermal layer of onion scale
(c) Obtaining outer epidermal layer of onion scale
2. Mount specimen on slide
(a) Via wet mount
(b) Via smears [e.g. blood samples]
(c) Via hanging drop method [e.g. live samples]
4. Observe specimen - Use of Microscope
(a) Focusing to obtain a clear image
