Epigenetic modulators play a key role in regulating gene expression as well as DNA damage response. These modulators include histone modifying enzymes and ATP-dependent chromatin remodeling proteins.
We seek to understand:
Role of ATP-dependent chromatin remodeling proteins in Candida albicans: Candida albicans is an opportunistic fungus that causes Candidiasis. We are investigating the role of ATP-dependent chromatin remodeling proteins in the pathogenesis and virulence of the organism.
Regulating the regulators: How are the epigenetic regulators regulated? Our studies show that these epigenetic regulators, in fact, regulate each other. For example, we have shown that SMARCAL1, a distant member of the ATP-dependent chromatin remodeling protein family, and BRG1, another member of the family, mutually co-regulate each other in mammalian cells, and thereby regulate the DNA damage response pathway. In C. albicans, we find that, Rtt109 mediates H3K56 acetylation which is needed for Fun30 expression. In turn, Fun30, an ATP-dependent chromatin remodeler, regulates the transcription of Rtt109. Both together mediate DNA damage response pathway.
