IV. Acute slice preparation tips

SCJ ver. acute slice cutting solution

pH 7.3~7.4

• This solution is used only for cutting acute slices.
  After cutting, incubate and store slices in normal recording solution.
  
  

• This high-Mg²⁺ solution works well for brains from young rats or mice (up to 4 weeks old).
  For older animals, sucrose-based solutions may be preferable
  (Dr. Bunano lab protocol; Kwon et al., 2008, PNAS).
  
  

• Sagittal sections are generally used for somatic recordings, and semi-sagittal sections for dendritic patches.
  
  

• A slice thickness of 350 µm is suitable for EPSP and EPSC recordings.
  
  

• (SCJ ver.) Incubation at 37 °C for 30 min after cutting was optimal for EPSC recordings.
