Yes, you do, but it doesn't always have to be a teaching degree.
You must have a university degree in order to receive an instructor visa (that lets you live and work in Japan as a teacher).
Working Holiday makers don't need a university degree to teach at English conversation schools.
If you want to teach on the JET Programme (below), then you don't need to have studied teaching. Many people try teaching on the JET Programme before starting other careers.
If you are interested in being a full-time teacher in a Japanese junior or senior high school, then you should consider a degree in Education and/or TESOL (Teaching English to Speakers of Other Languages).
Participants of the JET Programme primarily work as Assistant Language Teachers (ALTs), contributing to English education in schools and facilitating cross-cultural exchange. While it is a competitive program, accommodation and travel expenses are supported.
The JET Programme also has the following positions:
Coordinator for International Relations (CIR): Work in local Japanese government, prompting cultural exchange and doing translation and interpretation.
Sports Exchange Advisors (SEA): SEAs are involved in promoting sports and physical education in schools and local communities.
