Magnetic nanoparticles (m-Torquers)
Small size (150 – 500 nm)
Biocompatible & non-toxic: silica coated, PEGylated
0.1 – 50 pN force generation (adequate for most mechanosensitive receptors)
MG instrumentation (Magnetic Force Generator, MFG)
Long working distance (~1 meter: similar to MRI bore size)
Scalable: 16-, 35-, and 70-cm diameter à larger setup is possible
Stimulation frequency: up to 10 Hz (permanent magnet), ~30 Hz (electromagnet)
Mechanosensitive proteins
Versatile platform for diverse mechanosensitive ion channels & receptors
Ion channels: (for excitation) Piezo1/2, TRP family, (for inhibition) FLYC1
Receptors: T cell receptor, Notch (synNotch), integrin,…
Delivery: viral (AAV, Lenti, AV), non-viral (LNP)
Target molecules:
Our MG is the versatile platform for any in vivo mechanobiology systems
Mechanosensitive ion channels (Piezo1/2, TRP, FLYC…)
Mechanosensitive receptors (T cell receptor, Notch/synNotch, Integrin…)
Molecular specificity is achieved by nanoparticles selectively binding and activating target mechanoreceptors via antigen-antibody interaction
Cellular specificity is achieved by selective genetic encoding of mechanosensitive ion channels in any target neuronal types
- genetic: neuron-type specific promoter, Cre-loxP, transgenic animal
- non-genetic: tissue-specific virus
Temporal control:
With current rotating magnetic field setup, continuous or pulsed (up to 10 Hz) stimulation is possible.
New electromagnetic setups being developed for >30 Hz stimulation
Fast cellular response:
Direct force delivery and activation of ion channels
Nearly instantaneous activation & deactivation depending on ion channel gating kinetics (<10 ms)
