Biochemistry - microbiome
BIOCHEMICAL ANALYSES
Sampling for biochemical analyses in the framework of COST Seawheat is coordinated by Ronan Sulpice (Univ. Galway). For the analyses roughly 100 mg freeze dried material per sample (specimen) is needed. The material MUST be originating from same specimen you would have sent for genetic analyses and eventually microbiome analyses. If you plan to make all three analyses (genotyping, microbiome and biochemical analyses). Microbial and genotyping require 1-2 cm2, so we would take the remaining of the specimen for biochemical analyses.
Please ensure the specimens collected are labelled the same way for genotyping and biochemical analyses. For biochemical analyses: I would suggest that you take at least 5 samples per site/seasonal timepoint, especially if the area is rich in Ulva species.
After collecting the specimen, place them in tubes filled with seawater and bring them back ASAP to the laboratory in a cool box if possible. Freeze your samples as fast as possible and store at -20 prior freeze drying.
Ideally we would like to receive the samples in 2 ml microtubes.

MICROBIOME ANALYSES
Sampling for biochemical analyses in the framework of COST Seawheat is coordinated by Aschwin Engelen (Univ. Algarve, Faro). The sampling for the microbiome requires 1-2 cm2 of material. The same procedures apply as for diversity sampling but the tissue is stored in Longmire buffer. Avoid touching the tissue with non-sterile surfaces (incl. hands - wear gloves). Do not wipe the tissue clean, as this will remove the microbiome.
