Up-B Basics
Up-b has three types of throws:
Prespike (opponent is less then 50%)
Spike (opponent is above 50%)
Grounded (opponent's feet are on the ground when thrown. this is not the scoop hitbox!)
Prespike Throw Common Combos
all of these should generally be performed after double-jumping. All of these are percent, rage, and character specific but will usually work on the said DI.

Spike Throw Combos
Spike throw combos are very positioning dependent.
If there are no plats, we cannot follow up if the opponent DI's away, unless near ledge so that if the opponent DI's away they get spiked to their death. We can follow up with jump dair, uair, fair/bair, or side-b depending on %. We have to read DI for these. Here's a video on a spike throw killing 50/50, but do note that due to buffs in 11.0.0, this killing 50/50 works around 10-15% earlier then is listed here.
Grounded Throw
Grounded throw is really complex and has not been labbed out fully. This depends heavily on character. Generally, it's best to try to hit the opponent with a down air after this.
