我々の脳では、多様な神経細胞がコミュニケーションをとり、高度な情報処理が行われています。神経細胞は、情報を受け取ると、自身の特性を変化させて環境に柔軟に適応していきます。また、神経回路が形成される発達期においても細胞間コミュニケーションは重要です。私たちの研究室では、膜電位を介したコミュニケーションに着目し、神経回路が形成され、機能するメカニズムおよび、基礎研究の立場から疾患の理解を深める研究を行っています。
The brain performs sophisticated information processing through communication among diverse types of neurons. In response to incoming signals, neurons dynamically modify their properties, enabling them to adapt flexibly to their environment. Cell-cell communication is also essential during brain development, when neural circuits are established. Our laboratory focuses on membrane potential-mediated communication to uncover the mechanisms underlying the formation and function of neural circuits. Through basic research, we also seek to advance our understanding of the cellular and molecular mechanisms of neurological disorders.
大脳皮質は細胞分裂、分化、移動樹状突起伸長、軸索投射およびシナプス形成といった複雑な過程を経て神経回路が形成されます。これらの過程が進む際、周囲の細胞あるいは環境からどのような相互作用をし、それが細胞内シグナルを駆動し、神経発生に寄与するか、いまだ明らかではありません。私たちはこれまでに、移動中の細胞では自発活動及び細胞内カルシウムシグナルが低く、自発活動の上昇が大脳新皮質神経細胞の移動の停止及び神経細胞の成熟を促すことを報告しました(Bando et al., Cereb. Cortex, 2016)。現在、細胞内カルシウムシグナルがどのように遺伝子発現を調節し、神経回路形成に寄与するか調べています。また、研究対象を海馬にも広げ、細胞間相互作用及び細胞内シグナルによる神経回路形成機構を研究しています。
Neural circuits in the cerebral cortex are formed through complex processes, including cell division, differentiation, migration, dendritic elongation, axonal projection, and synapse formation. However, it remains unclear how interactions with surrounding cells or the environment during these processes drive intracellular signaling and contribute to cortical development. We have previously demonstrated that migrating neurons exhibit low levels of spontaneous activity and intracellular calcium signaling, and that an increase in spontaneous activity promotes the termination of neuronal migration and the maturation of neocortical neurons (Bando et al., Cerebral Cortex, 2016). We are currently investigating how intracellular calcium signaling regulates gene expression to control neural circuit formation. In addition, we have expanded our research to the hippocampus, where we study the mechanisms underlying neural circuit formation through cell-cell interactions and intracellular signaling.
神経細胞及び回路機能を理解するためには、高い時空間分解能で、長期間活動を記録、解析する技術が不可欠です。私たちは膜電位の光学記録に特に着目し、これまでにシナプス入力を高感度に検出できる蛍光プローブ・ArcLight-ST及びKv-ArcLight-STを開発しました(Bando et al., Cell Rep., 2019; Nat. Commun., 2021)。現在、蛍光膜電位プローブの更なる改良と応用を進めています。
Understanding the function of neurons and neural circuits requires technologies capable of recording and analyzing neuronal activity over extended periods with high spatiotemporal resolution. We have focused on the optical recording of membrane potential and developed the genetically encoded fluorescent voltage indicators ArcLight-ST and Kv-ArcLight-ST, which enable highly sensitive detection of synaptic inputs (Bando et al., Cell Reports, 2019; Nature Communications, 2021). We are currently improving fluorescent voltage indicators.
神経回路形成不全はしばしば精神・神経疾患につながることが知られています。私たちはこれまでに、重篤な精神遅滞をきたすヒト遺伝性疾患、Birk Barel症候群の原因遺伝子であるKCNK9の発現/機能抑制により、大脳皮質神経細胞移動が障害されることを明らかにしました(Bando et al., Cereb. Cortex, 2014)。当研究室では、神経回路形成の異常がどのように疾患を引き起こすか、細胞、回路、個体レベルで階層横断的に解析しています。
Defects in neural circuit formation are often associated with neurodevelopmental and neuropsychiatric disorders. We previously demonstrated that suppression of the expression or function of KCNK9, the causative gene for Birk-Barel syndrome, a hereditary disorder characterized by severe intellectual disability, disrupts the migration of cortical neurons (Bando et al., Cerebral Cortex, 2014). Our laboratory investigates how abnormalities in neural circuit formation lead to neurological and psychiatric disorders through an integrated, multiscale approach spanning the cellular, circuit, and whole-animal levels.
