サンプル：マウスE14.5肺 2ml tubeを使用

100％メタノールに回収

　　↓ r.t. ３hr

新しい100%メタノールに交換し -20℃保存

メタノールからPBSに置換

　　↓

2mm角にきざむ

1.5% Normal Rabbit Serum 200μl

　　↓ r.t.1hr 回転振盪

１次抗体 anti- Shh x10 inPBS 200μl

　　↓ 4℃ o/n

Wash PBS

　　↓ r.t. 回転振盪10min x3

2次抗体 anti-goat-Alexa x200 inPBS 200μl

　　↓ 4℃ o/n

Wash PBS

　　↓ r.t. 10min x5遮光

コンフォーカル観察

　　↓

透明化液（Scale）に交換

　　↓ r.t. 4day回転振盪 遮光
