Early reports claimed the story was about humans who tried to lure Predators with Alien eggs, although the idea was scrapped.[24] Influenced by the work of Erich von Dniken, Anderson researched von Dniken's theories on how he believed early civilizations were able to construct massive pyramids with the help of aliens, an idea long debunked and based on misinterpretations of Aztec mythology.[25] Anderson wove these ideas into Alien vs. Predator, describing a scenario in which Predators taught ancient humans to build pyramids and used Earth for rite of passage rituals every 100 years in which they would hunt Aliens. To explain how these ancient civilisations "disappeared without a trace", Anderson came up with the idea that the Predators, if overwhelmed by the Aliens, would use their self-destruct weapons to kill everything in the area.[25] H. P. Lovecraft's novella At the Mountains of Madness (1931) served as an inspiration for the film, and several elements of the Aliens vs. Predator comic series were included.[19][26] Anderson's initial script called for five Predators to appear in the film, although the number was later reduced to three.[25]

In 1994, Atari Corporation released the Rebellion Developments-developed first-person shooter Alien vs Predator for the Atari Jaguar, in which one could play as an Alien, Marine, or Predator. On October 28, 2010, io9 published an exclusive interview with the Brothers Strause in which they revealed that Alien vs. Predator 3 would have led directly into Alien. Greg Strause stated that, "The original ending for AVPR, that we pitched them, ended up on the Alien homeward [sic], and actually going from the Predator gun, that you see at the end, it was going to transition from that gun to a logo of a Weyland-Yutani spaceship that was heading to an alien planet. And then we were actually going to cut down to the surface [of the alien planet] and you were going to see a hunt going on. Quick Take: A 2010 refresh of Rebellion's original game from 1999, Aliens versus Predator offers a new and exciting action-packed installment. There is an individual campaign for each playable race: Aliens, Colonial Marines, and Predator. The Colonial Marine missions provide plenty of entertainment and excitement, but the fun falls flat once you get to the other campaigns. Graphics look fresh for being ten years old but beware of map fatigue as the designers reuse the same maps and layouts throughout the game. Awkward controls and clumsy surface transitioning hurt the Alien and Predator campaigns. Sound design and audio mix seem mostly great, however, a few voice-acted moments within the Marine audio diaries are quite amateurish. No online multiplayer matches to be found. It's a shame, but common for many games even only five years old.

Marine Campaign: The Marine missions are fast-paced and play similarly to modern DOOM games. Constant gunfire and fast-paced movement. Also similarly, It doesn't impart terrifying or horrific emotions outside of a few jump scares. It may be creepy or horror elements and themes, but it's not done in a particular or even purposefully scary way. Aliens do flock at you while you spam bullets at them, which can be a bit emotionally overwhelming at first - I'll admit! Nonetheless, it makes for a fun romp around shooting things. Fights against the Predator are way cooler and more fulfilling than the previous AVP game. The improvement in graphics, AI, and atmospheric particle effects like laser beams and smoke/fog. It makes for a more involved and exciting experience. Disappointed that the Marine campaign is almost entirely player versus Aliens. The Predators' appearance is just shy of being a cameo. The overall story for the Marines is nothing too special, but fit in well enough to keep the momentum going.

An unofficial port to Linux was later released in 2001, following Rebellion's public release of the game's source code. The game features three separate storylines for each of the playable factions, the Aliens, the Predators and the Colonial Marines; while each plot is unconnected, the Marine story loosely follows on from events of Alien and Aliens, while the Predator plot includes elements taken from Alien3.

The game is often considered the successor to the Alien vs Predator game on the Atari Jaguar, and as such is the second game in the Alien vs. Predator franchise. It was followed by two sequels, Aliens versus Predator 2 (2001) and Aliens vs. Predator (2010).

As a Predator, the player may make use of Predator weapons such as the Plasmacaster, Smart Disc, Wristblades and the Speargun capable of tearing off body parts and embedding them in walls. Predators are also considerably more athletic than humans, are the most durable creature in the game by far (even surpassing the armor capacity of armored humans in terms of withstanding damage), can make use of a cloaking device to stalk prey, and have a range of different vision modes to aid them in hunting the other species; in addition, they are capable of falling from far greater heights than humans without fear of injury or death. In the game, default Predator vision is similar to that of humans, but an infrared vision mode is available for tracking humans (similar to that in the Predator films), while a vision mode sensitive to electrical systems allows Predators to track Aliens (and android). Playing as an Alien is the most significantly different perspective. Players are able to range freely over any surface regardless of its inclination. However, weaponry is restricted to claws, tail and inner jaws (for the infamous Headbite), though these themselves reward the player with a fairly unusual experience in FPS combat. By default, Aliens visually perceive the environment in a similar manner to humans, albeit distorted by a fisheye lens to increase field of view; it is possible this view style was influenced by Alien3, which includes footage "seen" from the Xenomorph's point of view filmed using a fisheye lens. Players can also switch to a "sonic resonance" or echolocation mode to increase visibility in dark environments (as explained in Aliens versus Predator 2). This is augmented by an ability to detect pheromones and to discern human or Predator prey. The Aliens in this game can drop from any height without fear of injury or falling, and are the fastest of the three character species that the player can use.

Aliens versus Predator Gold Edition was released in 2000, approximately one year after the original game. This version adds two new weapons for the Marines (the Pistol and the Skeeter Launcher), the ability to save progress in singleplayer (although players have limited saves depending on the level difficulty) and numerous new multiplayer levels. The in-game video messages (seen on video screens throughout the singleplayer campaigns) were also replaced with newly filmed versions, featuring members of Rebellion's staff instead of the hired actors originally used.
