Disease resistance is a key trait in the development of improved crop varieties and high-quality seeds. Plant defense against pathogens involves complex interactions among multiple genes. While previous studies have primarily focused on model plant–pathogen systems, recent advances in next-generation sequencing and bioinformatics have enabled large-scale identification of disease resistance-related genes in diverse crops. Building on these advances, gain-of-function and loss-of-function studies are now being used to investigate the roles of individual genes, providing valuable insights into plant defense mechanisms and supporting the development of disease-resistant crops.
(Genomic and Big Data Analysis of Plant–Microbe Interactions)
Disease-resistant plants defend themselves against pathogens by regulating the expression of various genes. Understanding these defense mechanisms requires a comprehensive analysis of plant–pathogen interactions at the molecular level. Recent advances in next-generation sequencing (NGS), particularly RNA sequencing (RNA-seq), have enabled large-scale analysis of gene expression changes during pathogen infection. Our laboratory uses comparative transcriptomic analysis to investigate plant–pathogen interactions, identify genes involved in disease resistance, and elucidate the molecular mechanisms underlying plant immunity.
(Identification and Characterization of Plant Genes Involved in Disease Resistance)
Plant resistance or susceptibility to pathogens is determined by complex interactions between plants and invading microorganisms. While pathogens produce virulence factors to promote infection, plants have evolved diverse defense mechanisms, including resistance (R) proteins that recognize pathogen effectors and activate immune responses. Identifying these pathogen-derived factors and their interacting host proteins is essential for understanding plant immunity.
Our laboratory uses plant virus-based expression vectors to produce epitope-tagged recombinant proteins in plants, followed by immunoprecipitation and liquid chromatography–tandem mass spectrometry (LC–MS/MS) to identify interacting host proteins. Through these approaches, including studies of Soybean mosaic virus (SMV), we aim to identify host proteins that interact with viral pathogenicity factors and characterize protein interaction networks involved in regulating plant disease resistance and susceptibility.
(Development of Disease-Resistant Crops)
Plant viruses are disease-causing pathogens, but their small genomes and ability to spread systemically within plants make them valuable tools for studying gene functions. By genetically modifying plant viruses into gene expression vectors, researchers can introduce and express target genes throughout infected plants, enabling gain-of-function studies.
Plants also possess a natural defense mechanism called RNA silencing, which recognizes and suppresses foreign RNA, including viral RNA. This mechanism can be exploited through virus-induced gene silencing (VIGS), in which plant virus vectors carrying sequences derived from host genes are used to suppress the expression of specific plant genes. VIGS therefore provides an effective tool for systemic loss-of-function studies.
Our laboratory develops and applies plant virus-based gene expression and silencing technologies to investigate the functions of genes associated with desirable crop traits, with a particular focus on identifying disease resistance-related genes and developing disease-resistant crop varieties.
(Development of Plant Virus-Based Gene Expression Technologies)
Plant pathogens cause an estimated 5–20% loss in crop productivity worldwide, while climate change and increasing international trade have heightened the risk of emerging and unexpected disease outbreaks. Although conventional breeding has produced numerous disease-resistant crop varieties, the genes and molecular mechanisms underlying disease resistance remain incompletely understood. Furthermore, the emergence of new pathogen strains and invasive pathogens poses growing threats to existing resistant varieties.
These challenges highlight the need to develop crop varieties with durable disease resistance through advanced molecular breeding approaches. Our laboratory investigates the molecular mechanisms of disease resistance in major crops, including soybean, and applies these findings to molecular breeding and advanced seed biotechnology. Our ultimate goal is to develop crop varieties with improved resistance to plant pathogens, contributing to sustainable agricultural production.
