1. Protein and enzyme purification and their biochemical characterization: Protein extraction (total, apoplastic, nuclear and plasma membrane), size exclusion chromatography, ion exchange chromatography, affinity chromatography, western blotting, protein purification, protein quantification etc.
2. Analysis of “Hidden Proteome”: Enrichment of low-abundance proteins using various biochemical techniques and precipitation methods (protamine sulfate and ethanol precipitation methods).
3. Post-translational modifications analysis: Isolation, enrichment, characterization and mass spectrometry-based identification of phosphorylated and glycosylated proteins.
4. Enzymatic activity analysis: Spectrophotometric and gel-based enzymatic activity analysis. Analysis of antifreeze activity using nanoliter osmometry and splat assays.
5. Gel-based Protein Separation Technologies: One and two-dimensional gel electrophoresis (1- and 2-DGE), Differential in-gel electrophoresis (DIGE) and Native PAGE. Image acquisition of gel using scanner such as Typhoon 9410 variable mode imager followed by down-stream analysis using ImageMaster2DPlatinum software and PDQuest software.
6. Mass Spectrometry: Experience in in-gel and in-solution trypsin digestion (by both conventional and FASP methods), TMT-labelling and enrichment of phosphopeptides, peptides desalting, SCX and RP fractionation of peptides and basics of MALDI-TOF operation.
7. Isolation and quantification of metabolites: Isolation and HPLC based quantification of metabolites including isoflavonoids, amino acids, and ascorbic acid among others.
8. Bioinformatics and Statistical analysis: Mass spectrometry acquired data processing using Mascot and other software. Label-free quantitation of MS obtained raw data by MaxQuant and downstream data processing by Perseus software.
