Commentary
My construction of this family is based on DNA evidence, naming patterns, and the apparently shifting home base of William and Elizabeth, which helps cement identifications.
Fairly good DNA evidence ties descendants of the John Sadler who was buried in 1815 in Osmotherly and of the Wilfred Sadler who was buried in 1809 in Leake. Given the clustering and the amount of DNA involved, it seems likely that John and Wilfred were brothers.
John’s burial entry in the parish register records his age as 88. By far the most plausible baptism in the area was in Kirby Sigston in 1727. The father in this baptism is recorded as William.
A Wilfred Sadler was baptized in Northallerton in 1721, with the father recorded as William. I will argue that this Wilfred and John were brothers. (Before going further, I will note that several websites have Wilfred being baptized in Alne in 1720. This is an error. That Wilfred died as an infant.)
William Sadler and Elizabeth Kilton married in Northallerton in 1717. Elizabeth was probably from Kirby Sigston. (A Kilton family lived in Kirby Sigston, and a James Kilton had a marriage recorded in both the Northallerton and Kirby Sigston registers in 1719. Northallerton and Kirby Sigston are about two miles apart.) The Northallerton and Kirby Sigston registers in the period name only the fathers in baptisms (and some burials). Here are entries with the father recorded as William Sadler:

It seems reasonable to me to think that this is one family. There is no overlap in time, Northallerton and Kirby Sigston were very close, and the William buried in 1730 is plausibly the one baptized in 1718.
By the late 1740s, William and Elizabeth appear to have moved to South Cowton, about 10 miles away. The South Cowton register records the burial in 1747 of Frances, daughter of William and Elizabeth Sadler. The Frances baptized in Kirby Sigston in 1732 is the only plausible candidate for this burial. In 1748 in South Cowton, Elizabeth Sadler married John Kitchen. John and Elizabeth named their eldest child Frances. It seems likely that this is the Elizabeth baptized in Northallerton in 1720, and that she named her first child after her recently deceased sister. In 1755 in South Cowton John Sadler and his wife Susannah had a daughter Frances baptized. This was the John Sadler who was buried in Osmotherley in 1815, and Frances was his eldest child.
Overall, I think the evidence tying this family together is reasonably good.
