Program 1. Vancomycin Heptapeptides and Derivatives

The first area of interest is the development of unnatural glycopeptides with antimicrobial activity. Antibiotic resistance is a major public health concern and the occurrence of drug-resistant, disease-causing bacteria has become more acute over the past six decades. The aim of our first study is to develop a chemoenzymatic synthesis of vancomycin and novel derivatives. This research will serve as a platform for the design of new antimicrobial glycopeptides.

Program 2. Aryl triflates as precursors to Morphine Derivatives

The second research program aims to address the opioid epidemic. The use of morphine for pain relief has led to misuse and drug dependence. The fluorine atom has been shown to improve lipophilicity, improve potency, lower basicity of nearby nitrogen atoms, and increase hydrogen bonding. Owing to the small size of fluorine, all of this is possible while maintaining the binding (activity) of the parent compound. These fluorinated morphines are hypothesized to be important tools for manipulation of morphine’s polypharmacology, warranted by its importance in pain treatment, but contrasted by its drawbacks: addiction, patient tolerance, and withdrawal.
