Lab 7: Mitosis & Meiosis
Objectives:
Describe the events in the cell cycle and how these events are controlled.
Explain how DNA is transmitted to the next generation via mitosis.
Explain how DNA is transmitted to the next generation via meiosis followed by fertilization.
Understand how meiosis and crossing over leads to increased genetic diversity, which is necessary for evolution.
Things to note in this lab:
Make sure you answer the questions for each section of the lab (if called for).
Everything will be in the lab notebook.
DO NOT WRITE ON A SEPARATE SHEET OF PAPER.
This lab will be broken down into
2 parts
. It is also an
INDIVIDUAL
lab!
We will be collecting class data for part 2. Please enter in your data so your fellow classmates can complete their lab as well.
For parts 1 & 4, you will
not
draw them in your notebook but show me each step as you complete it in class.
Part 1 Procedures:
Using pop-beads or chalk, illustrate the stages of interphase and mitosis.
Answer questions along the way as given.
Part 2 Procedures:
Prepare an onion root tip slide.
Place an onion root tip in 1M HCl for 4 min.
Transfer to Carnoy's fixative for 4 min.
Remove a slide from jar with 70% ethanol. Dry with wipe & label with wet erase marker.
Place tip on slide & cut off distal 2mm portion of tip. Discard the remainder.
Cover root tip with carbol-fuschin stain for 2 min.
Blot off excess & cover tip with 1-2 drops of water.
Gently tease tip apart with probe. Place cover slip over root tip & clean.
Firmly press down with eraser of a pencil.
Don't break cover slip by pressing too hard
.
Count cell at high magnification. Use well stained cells. Count those in view at each phase. Repeat in 2 other root tips.
Collect the class data for each group and calculate the mean & standard deviation for each group.
Compare the number of cells from each group.
Collect class data & enter into a table. These are the observed values.
Use the data to calculate the totals with the formulas in the manual.
Use the totals from above to calculate the expected values using formula in manual.
Enter the observed values & expected values in table 4. Calculate the chi-square value.
Compare chi-square to critical value.
Part 3 Procedures:
Answer the pre-lab questions.
Go through Case 1 on HeLa cells and research & answer questions.
Go through Case 2 on Philadelphia chromosomes and research & answer questions.
Part 4 Procedures:
Using pop-beads or chalk, illustrate the stages of interphase, meiosis I & meiosis II.
Answer questions along the way as given.
Part 5 Procedures:
Place a drop of water on a slide.
Scrape some cells from neat the 2 strains meeting.
Place a cover slip over the cells.
Gently press down on slip.
Count at least 50 ASCI & count as parental or recombinant.
Enter data & make calculations.
parent recombinant = 1 map unit
% ASCI show recombinant divided by 2 = map units separating spore-color gene from centromere
% ASCI show recombinant divided by 2 because only half of spores in each are a result of crossing-over.
